Proof by chemical equation why six mols of oxygen are required to oxidise 12 molecule of glucose

C6H12O6 + 6O2 --> 6CO2 + 6H2O

If I read your statement correctly, and assuming you're using the same equation as above, no, I don't believe it.
6 moles O2 is required to oxidize 1 mole glucose; however, 1 mole of glucose is much much much more than 6 molecules.
