1) Explain the purpose of the controls used in the mating experiment
2) It is necessary to make dilutions prior to plating in Week 1 in order to obtain single colonies. Why are single colonies important in this experiment?

I can't help on the first, because I don't know what the experiment is.
2. One makes dilutions to get a very low bacteria count...with that, you will not have multiple colonies on the plates. AFter all, you are doing a "mating" experiment and examining genetic heredity. If you were doing dogs, you would not use multiple males with one female for obvious reasons, one could never know where the genetic material came from.
